I currently download and upload all data from my computer to my MX-850 remote via the supplied serial port cable. I am purchasing a new computer. Does the new computer need a serial port, or is there a cable that will reliably interface the serial port pin connector cable with a USB connection to communicate with the remote?

You will need to get a USB-Serial adaptor. Do a search on here, there are a few that will work fine and alot more that won't work.

If your new laptop has an expresscard slot, you can get one of these off ebay. I got one and it works fine with my mx850.
