You need to first make sure you are not just creating a graphic which contains an empty layer. If you are that's the problem. The layer has to consist of a solid color, preferably white with an opacity setting of 1%, and saved as a PNG-24. If you are following these steps the resulting button will be clickable in both the Simulator and remote.

The transparent images in the gallery you speak of are text panels which are completely transparent. They were not saved with the 1% opacity required, so that they would be clickable if used as buttons.
